摘要:Proteins that transmit molecules and signals across the plasma membrane are crucial in cell biology because they enable cells to sense and respond to their surroundings. A major challenge for studying cell surface proteins is that often they do not fold or traffic properly to the plasma membrane when produced in heterologous cells. We developed a strategy for quantifying surface localization from fluorescence microscopy images of surface-stained cells. Using clustered protocadherins, a protein...

摘要:HvAACT1 is a major aluminum (Al)-tolerance gene in barley, encoding a citrate transporter that belongs to the multidrug and toxic compound extrusion (MATE) family. This transporter facilitates citrate secretion from the roots, thereby detoxifying external Al ions-a major constraint of crop production on acidic soils. In this study, we present the outward-facing crystal structure of HvAACT1, providing insights into a citrate transport mechanism. The putative citrate binding site consists of thr...

摘要:Emerging infectious diseases increasingly threaten many wildlife populations, yet the impacts of pathogens vary considerably both within and among host species. The environmental tolerance mismatch hypothesis (ETMH) suggests that this variability stems in part from differences in the relative performance of hosts and pathogens under varying environmental conditions. According to the ETMH, pathogen impacts should be more severe in environments where pathogen performance is high and host perform...

摘要:While somatic variants are well-characterized drivers of tumor evolution, their influence on cellular fitness in nonmalignant contexts remains understudied. We identified a mosaic synonymous variant (m.7076A > G) in the mitochondrial DNA (mtDNA)-encoded cytochrome c-oxidase subunit 1 (MT-CO1, p.Gly391=), present at homoplasmy in 47% of immune cells from a healthy donor. Single-cell multiomics revealed strong, lineage-specific selection against the m.7076G allele in CD8+ effector memory T cells...

摘要:Land- use changes are reshaping the distribution of aboveground species worldwide. However, the impact of land- use changes on the distribution of soil organisms remains poorly understood. In particular, we lack a mechanistic understanding of the environmental factors reshaping the distribution of soil microbiota in response to global biological homogenization. Here, we used metabarcoding to investigate the biogeography of protists and their relationships with prey and hosts in three human- do...

摘要:Nature- based solutions (NBS) are used to transform existing unsustainable and undesirable path dependencies in cities. For NBS to contribute to just urban transformations, a stronger inter- and transdisciplinary knowledge base is needed. This knowledge base is essential to engage with six complex yet crucial questions about NBS, including for what?, which nature?, where?, how?, when, and for whom?. To address these questions, we identify two critical opportunities to advance the knowledge of ...

摘要:G protein-coupled receptors (GPCRs) play pivotal roles in cellular signaling and represent prominent drug targets. Structural elucidation of GPCRs is crucial for drug discovery efforts. However, structural studies of GPCRs remain challenging, particularly for inactive-state structures, which often require extensive protein engineering. Here, we present a de novo design strategy termed click fusion for generating fusion proteins to facilitate GPCR structural studies. Our method involves the rat...

摘要:Theories of adaptive radiation propose predictable trajectories in which diversity accumulates rapidly in newly formed or colonized environments with underexploited niche space and few competing species, before slowing down as competition intensifies, and speciation and extinction rates approach equilibrium. This historical perspective on diversity may be more important than current environmental variation for explaining today's biodiversity, but this has been difficult to determine because of...

摘要:The Sterile Insect Technique (SIT) is a species-specific and environmentally friendly method for effectively controlling pest insect populations based on releasing reared, sterile insects into infested areas. Sex sorting in rearing facilities, enabling male-only releases, is necessary to ensure SIT programs are efficient, cost-effective and, in case of mosquito control, also safe. This can be greatly facilitated by genetic sexing strains (GSS), exhibiting sex-specific phenotypic markers. Howev...
