Let Y(x) be the toxicity associated with some [drug] dosage level x. Let the conditional distribution of Y be normal with mean linear in x. The optimal dosage is defined to be the largest value of x such that the probability that Y(x) does not exceed .eta. is at least .gamma., where .eta. is some threshold toxicity level and .gamma. is some specified tolerance probability. Recently, Eichhorn and Zacks have introduced the problem of sequentially and consistently estimating the optimal dosage at the same time controlling the probability that a dose does exceed the optimal dose. Sequential search procedures are defined here which control the probability that the toxicity exceeds .eta., the threshold value. Two models are considered corresponding to 2 different variance functions.
